Unconventional Antibacterials and Adjuvants

چکیده
The need for new classes of antibacterials is genuine in light the dearth clinical options treatment bacterial infections. prodigious discoveries antibiotics during 1940s to 1970s, a period wistfully referred as Golden Age Antibiotics, have not kept up face emergence resistant bacteria past few decades. There has been renewed interest old drugs, repurposing existing and pairing synergistic or an antibiotic with adjuvant. Notwithstanding, novel these life-saving drugs become increasingly difficult, calling paradigms. We describe, herein, three strategies from our laboratories toward adjuvants using computational multidisciplinary experimental methods. One approach targets penicillin-binding proteins (PBPs), biosynthetic enzymes cell-wall peptidoglycan, non-?-lactam inhibitors. Oxadiazoles quinazolinones emerged two structural out efforts. Several hundred analogs synthesized fully characterized laboratories. A second ventures into inhibition allosteric regulation biosynthesis. mechanistic details PBP2a Staphylococcus aureus, discovered laboratories, outlined. site this protein at 60 distance active site, whereby ligand binding former makes access latter by substrate possible. documented that both ceftaroline, fifth-generation cephalosporin, bind manifestation antibacterial activity. Bacteriophage lysins as antibacterials
We read the editorial entitled “Non-antibiotic treatments for bacterial diseases in an era of progressive antibiotic resistance” by Steven M. Opal published in Critical Care [1]. We want to congratulate the author for this timely editorial and appreciate the efforts to highlight various non-antibiotic-based approaches being pursued for treating drug-resistant bacteria. We fully agree that there...
متن کاملBacteriophages: antibacterials with a future?
The hypothesis as to whether a benign species of bacteria could kill a virulent kind has to this point been untested. Recently it was shown that in the macrophage, bacteriophages, when properly introduced through a nonvirulent microbe, had a killing rate for virulent AIDS Mycobacterium tuberculosis and Mycobacterium avium far in excess of modern day antibiotics. The study in effect brought a na...
متن کامل‘Evolution-Proofing’ Antibacterials
When antibiotics first came into use, they were so effective at curbing bacterial infections that it seemed the age-old battle of man vs. microbe would soon be at an end [1]. Eighty years and dozens of drugs later, we now know better. Following each new antibiotic’s launch, reports soon accumulated that once-treatable infections were becoming refractory to the drug [2]. Nowadays, many infectiou...

متن کاملInnate immunity and adjuvants
Innate immunity was for a long time considered to be non-specific because the major function of this system is to digest pathogens and present antigens to the cells involved in acquired immunity. However, recent studies have shown that innate immunity is not non-specific, but is instead sufficiently specific to discriminate self from pathogens through evolutionarily conserved receptors, designa...
